Decorin Antibody Microarray Validation. Analysis of HuProt(TM) microarray containing more than 19,000 full-length human proteins using Decorin antibody (clone DCN/3521). These results demonstrate the foremost specificity of the microarray validated DCN/3521 mAb. Z- and S- score: The Z-score represents the strength of a signal that an antibody (in combination with a fluorescently-tagged anti-IgG secondary Ab) produces when binding to a particular protein on the HuProt(TM) array. Z-scores are described in units of standard deviations (SD's) above the mean value of all signals generated on that array. If the targets on the HuProt(TM) are arranged in descending order of the Z-score, the S-score is the difference (also in units of SD's) between the Z-scores. The S-score therefore represents the relative target specificity of an Ab to its intended target.

Western blot analysis of kidney and lung tissue lysates from human, mouse, rat, hamster, guinea pig, and rabbit using Decorin Antibody (clone DCN/3521) detects bands at approximately 90-120 kDa, consistent with glycosylated forms of Decorin (DCN). The observed variation in band intensity and migration reflects species-specific differences in glycosaminoglycan modification and glycosylation state of this extracellular matrix proteoglycan. The conserved detection across multiple species supports reliable recognition of DCN and its post-translationally modified forms.

Western blot analysis of human 1) heart, 2) kidney, and 3) lung tissue lysates using Decorin Antibody (clone DCN/3521) detects bands at approximately 90-120 kDa, consistent with glycosylated forms of Decorin (DCN). Decorin is a secreted extracellular matrix proteoglycan, and its migration at higher apparent molecular weights reflects glycosaminoglycan modification and variable glycosylation states across tissues. The banding pattern across multiple tissues supports reliable detection of DCN and its post-translationally modified forms.
