Cytokeratin CAM5.2 Antibody Human Breast Carcinoma IHC. Immunohistochemistry staining of FFPE human breast carcinoma tissue was performed with Cytokeratin CAM5.2 Antibody, also called Cytokeratin 7/8 Antibody, using clone CAM5.2. Heat-induced epitope retrieval was performed by boiling tissue sections in 10 mM Tris with 1 mM EDTA, pH 9, for 20 min followed by cooling. Strong cytokeratin staining is observed in the epithelial tumor structures, with comparatively little staining in the surrounding tissue. This Cytokeratin CAM5.2 Antibody demonstrates low molecular weight cytokeratin expression in human breast carcinoma.

Cytokeratin CAM5.2 Antibody / Cytokeratin 7/8 Antibody is a monoclonal antibody recognizing low molecular weight cytokeratins expressed by many simple and secretory epithelial cells. The CAM5.2 clone reacts primarily with cytokeratin 8 (CK8/KRT8), with weaker but distinct reactivity with cytokeratin 7 (CK7/KRT7). These intermediate filament proteins have molecular weights of approximately 52 and 48 kDa, respectively, and contribute to the structural organization and mechanical stability of epithelial cells.

CK8 is broadly expressed in simple epithelia and commonly forms intermediate filament networks with other type I keratins. CK7 has a more restricted epithelial distribution and is frequently studied as a marker of epithelial differentiation. The combined reactivity of CAM5.2 produces a characteristic low molecular weight cytokeratin staining profile across numerous epithelial tissues. A Cytokeratin CAM5.2 Antibody can therefore support studies of epithelial identity, differentiation and intermediate filament organization.

CAM5.2 staining is found in many normal secretory and simple epithelial tissues, including hepatocytes and renal tubular epithelium, while stratified squamous epithelium generally shows limited or absent reactivity. This distribution has made CAM5.2 useful for investigating epithelial-derived tumors, including hepatocellular and renal cell carcinomas. Some squamous cell carcinomas may show weak or absent staining, reflecting differences in cytokeratin expression among epithelial tumor types.

An important distinction is that CAM5.2 should not be considered a CK8/CK18-specific antibody. Although CAM5.2 has sometimes been described this way, characterization of the clone supports predominant CK8 recognition with weaker CK7 reactivity rather than CK18 recognition. This distinction is important when interpreting CAM5.2 staining or comparing it with antibodies specifically targeting CK8, CK18 or CK8/CK18 complexes.
