This antibody recognizes the D1 domain of an N-glycosylated glycoprotein of 120kDa with intra-chain disulfide bonds, identified as CD50 or ICAM3 (Leucocyte Workshop VI). It is the major ligand for LFA-1 (CD11a/CD18) and may have signaling role to increase adhesion. It is expressed on thymocytes and T lymphocytes and is resistant to treatment with phosphatidylinositol (PI) phospholipase C. This CD50 antibody is excellent for staining of formalin/paraffin tissues.
The concentration stated for each application is a general starting point. Variations in protocols, secondaries and substrates may require the CD50 antibody to be titered up or down for optimal performance.
1. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes.
2. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.
Stimulated human leukocytes were used as the immunogen for this CD50 antibody.
Store the CD50 antibody at 2-8oC (with azide) or aliquot and store at -20oC or colder (without azide).
