Immunohistochemistry analysis of CD27 Antibody Recombinant Rabbit MAb in human lymph node tissue. FFPE human lymph node demonstrates membranous HRP-DAB brown staining in scattered lymphoid cells within paracortical and follicular regions, consistent with CD27/TNFRSF7 expression on activated and memory lymphocytes, while the PBS negative control inset shows no specific staining. Antigen retrieval was performed by boiling tissue sections in pH 9 10mM Tris with 1mM EDTA for 20 minutes followed by cooling prior to incubation with CD27 antibody (clone LPFS2/8316R).

CD27 antibody recognizes CD27, a type I transmembrane glycoprotein and member of the tumor necrosis factor receptor superfamily encoded by the CD27 gene. CD27 is primarily localized to the plasma membrane of T lymphocytes, memory B cells, and subsets of natural killer cells, where it functions as a co-stimulatory receptor that promotes lymphocyte activation, survival, and differentiation. CD27 Antibody Recombinant Rabbit MAb supports research focused on adaptive immunity, lymphocyte maturation, and immune checkpoint signaling.

The CD27 gene is located on chromosome 12p13 and encodes a protein containing extracellular cysteine-rich domains characteristic of TNF receptor family members, a single transmembrane region, and a cytoplasmic tail that recruits TRAF adaptor proteins. Upon binding to its ligand CD70, CD27 triggers activation of NF-kappaB and related signaling pathways that enhance T-cell expansion, promote memory formation, and support B-cell differentiation. CD27 expression is tightly regulated during immune activation and is commonly used to distinguish naive, memory, and effector lymphocyte subsets.

In secondary lymphoid tissues such as tonsil and lymph node, CD27-positive cells are observed within T-cell zones and germinal center-associated compartments. CD27 is broadly expressed on naive and central memory T cells and on memory B cells, but expression may be reduced in terminally differentiated effector populations. Dysregulated CD27 signaling has been associated with chronic infection, autoimmunity, and certain lymphoid malignancies. The CD27-CD70 axis is actively investigated in immuno-oncology, as modulation of this pathway can enhance anti-tumor immune responses.
