CD137 antibody targets CD137, also known as 4-1BB, a member of the tumor necrosis factor receptor superfamily encoded by the TNFRSF9 gene. CD137 is a type I transmembrane costimulatory receptor that is minimally expressed on resting immune cells but is rapidly induced following cellular activation. CD137 antibody reagents are widely used to study immune activation due to the receptorâs tightly regulated expression and strong association with effector immune responses.
CD137 expression is most prominently observed on activated CD8+ T lymphocytes, with additional expression detected on activated CD4+ T cells, natural killer cells, dendritic cells, and subsets of monocytes. Upon ligand engagement, CD137 signaling promotes intracellular pathways that support cell survival, proliferation, and sustained cytokine production. These signaling events contribute to the expansion and persistence of effector and memory immune cell populations during immune responses.
As a costimulatory immune receptor, CD137 plays an important role in shaping adaptive immunity and regulating immune activation thresholds. Elevated CD137 expression has been reported in inflammatory conditions and within tumor-infiltrating lymphocyte populations, where it reflects immune cell activation within the tissue microenvironment. Antibody-based detection of CD137 is therefore frequently applied in immunology and immuno-oncology research to characterize activation status, immune cell subsets, and costimulatory signaling dynamics.
In addition to its role in acute immune activation, CD137 signaling has been implicated in immune regulation during chronic stimulation and immune exhaustion. Analysis using a 4-1BB antibody enables detailed assessment of costimulatory receptor expression across experimental systems, including lymphoid tissues and activated immune cell populations. 1. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.
A recombinant human CD137 protein fragment within amino acids 19-188 was used as the immunogen for the CD137/4-1BB antibody.
Store the CD137/4-1BB antibody at 2-8oC (with azide) or aliquot and store at -20oC or colder (without azide).
