CD117 Antibody C117/6347 Protein Microarray Validation. Protein microarray analysis of CD117 Antibody clone C117/6347 using a HuProt array containing more than 19,000 full-length human proteins. KIT produces the dominant signal, clearly separated from the next-highest array proteins. CD117 Antibody C117/6347 in Human GIST IHC. Immunohistochemistry staining of FFPE human gastrointestinal stromal tumor (GIST) tissue with CD117 Antibody clone C117/6347. HIER was performed by boiling tissue sections in pH 9 10mM Tris with 1mM EDTA for 20 min followed by cooling before testing. Strong, widespread brown immunoreactivity is visible throughout the tumor-cell population, with comparatively pale nuclei. The robust staining in this highly relevant CD117-positive tumor context demonstrates strong tissue performance of the c-Kit Monoclonal Antibody.

Under reducing conditions, prominent bands are visible near the expected approximately 50 kDa heavy-chain and 25 kDa light-chain positions. Under non-reducing conditions, the intact antibody migrates predominantly as a high-molecular-weight species near approximately 150 kDa. The expected change in migration between reducing and non-reducing conditions confirms the integrity and purity of the c-Kit Monoclonal Antibody preparation.

CD117 Antibody C117/6347 / c-Kit Monoclonal Antibody recognizes KIT, a receptor tyrosine kinase commonly known as CD117, c-Kit and the stem cell factor receptor. KIT belongs to the type III receptor tyrosine kinase family and consists of an extracellular ligand-binding region, a single transmembrane segment and an intracellular split kinase domain. Its ligand, stem cell factor, regulates KIT-dependent signaling in several cell populations involved in development, hematopoiesis and tissue homeostasis.

Binding of stem cell factor promotes KIT receptor dimerization and activation of its intracellular kinase domain. Subsequent receptor phosphorylation creates docking sites for signaling proteins that connect KIT with pathways including PI3K/AKT, RAS/MAPK and JAK/STAT signaling. Through these networks, KIT contributes to cellular proliferation, survival, differentiation and migration. Tight regulation of receptor abundance and kinase activity is therefore important for normal cellular function.

CD117 is an established marker associated with hematopoietic progenitor cells, mast cells, melanocytes and germ cells. KIT signaling is important for normal hematopoiesis, mast cell development, pigmentation and gametogenesis. The receptor is also widely studied in stem and progenitor cell biology, where CD117 expression is frequently used as part of phenotypic characterization strategies.

Altered KIT signaling is particularly important in cancer research. Activating KIT mutations and abnormal receptor signaling occur in gastrointestinal stromal tumors and subsets of other malignancies. CD117 expression is consequently investigated in studies of tumor biology and diagnostic marker expression, while KIT kinase activity is an established target of small-molecule inhibitors.
