Cadherin 17 Antibody Colon IHC. Immunohistochemistry staining of FFPE human colon tissue using Cadherin 17 Antibody / Liver-Intestine Cadherin Antibody (clone CDH17/2615) demonstrates strong membranous HRP-DAB brown staining throughout colonic epithelial cells lining glandular and crypt structures. The staining pattern is consistent with expression of CDH17 (LI-cadherin), a calcium-dependent cell adhesion molecule that contributes to epithelial organization, maintenance of tissue integrity, and regulation of cell-cell interactions within the gastrointestinal tract. CDH17 is highly expressed in intestinal epithelium and serves as a well-established marker of gastrointestinal epithelial differentiation and lineage. HIER was performed by boiling tissue sections in pH 9, 10 mM Tris with 1 mM EDTA for 10-20 minutes followed by cooling prior to immunostaining.
Analysis of HuProt(TM) microarray containing more than 19,000 full-length human proteins using Cadherin 17 antibody (clone CDH17/2615). These results demonstrate the foremost specificity of the CDH17/2615 mAb.
Z- and S- score: The Z-score represents the strength of a signal that an antibody (in combination with a fluorescently-tagged anti-IgG secondary Ab) produces when binding to a particular protein on the HuProt(TM) array. Z-scores are described in units of standard deviations (SD's) above the mean value of all signals generated on that array. If the targets on the HuProt(TM) are arranged in descending order of the Z-score, the S-score is the difference (also in units of SD's) between the Z-scores. The S-score therefore represents the relative target specificity of an Ab to its intended target.

Cadherin 17 (CDH17), also known as Liver-Intestine Cadherin (LI-cadherin), is a calcium-dependent cell adhesion molecule predominantly expressed in gastrointestinal epithelial tissues, where it contributes to epithelial organization, maintenance of tissue integrity, and regulation of cell-cell interactions. The observed molecular weight exceeds the predicted size of approximately 92 kDa, which is commonly attributed to glycosylation of the extracellular domain. Such post-translational modification frequently results in slower electrophoretic migration and an increased apparent molecular weight during western blot analysis. Predicted molecular weight: approximately 92 kDa.

Description

Cadherin 17 Antibody recognizes a protein of 120kDa, which is identified as Cadherin 17 (also known as LI Cadherin). The cadherins are a family of Calcium-dependent adhesion molecules that function to mediate cell-cell binding critical to the maintenance of tissue structure and morphogenesis. Cadherins each contain a large extracellular domain at the amino terminus, which is characterized by a series of five homologous repeats, the most distal of which is thought to be responsible for binding specificity. The relatively short carboxy terminal, intracellular domain interacts with a variety of cytoplasmic proteins, including beta-catenin, to regulate cadherin function. LI-cadherin (for liver-intestine-cadherin) expression is restricted to liver and intestine tissues and is specifically localized to the basolateral domain of hepatocytes and enterocytes.
