GTF2H2 Antibody / TFIIH p44 Antibody recognizes GTF2H2, the p44 subunit of general transcription factor IIH (TFIIH), a multiprotein complex with essential functions in RNA polymerase II transcription and nucleotide excision repair. GTF2H2 is a component of the TFIIH core complex and contributes to the molecular machinery responsible for transcription initiation and repair of damaged DNA. The protein is also historically known as BTF2 p44 or BTF2P44.
During RNA polymerase II-dependent transcription, TFIIH is recruited to the transcription preinitiation complex assembled at gene promoters. TFIIH participates in promoter opening and promoter escape, enabling RNA polymerase II to transition into productive transcription. GTF2H2 contributes to TFIIH function through interactions with other components of the complex, including the XPD helicase encoded by ERCC2. Its N-terminal region interacts with and regulates XPD, while the C-terminal portion contributes to successful RNA polymerase II promoter escape.
TFIIH also has a central role in nucleotide excision repair, a DNA repair pathway responsible for removing bulky, helix-distorting DNA lesions. The TFIIH core complex participates in both global genome and transcription-coupled nucleotide excision repair, helping open DNA surrounding a lesion so the damaged oligonucleotide can be excised and replaced. The dual involvement of GTF2H2 in transcription and DNA repair makes it relevant to studies of basal transcription, transcription factor assembly, genomic stability, DNA damage responses and nucleotide excision repair.
Clone PCRP-GTF2H2-1B9 is a mouse monoclonal antibody generated against full-length recombinant human GTF2H2 protein and purified by Protein A/G affinity chromatography. The antibody has also been evaluated on a HuProt protein microarray containing more than 19,000 full-length human proteins.
