Beta-2 Microglobulin Antibody C21.48A1 / Free B2M Marker detects beta-2 microglobulin (B2M), with clone C21.48A1 recognizing a hidden determinant that becomes available when beta-2 microglobulin is separated from the HLA heavy chain. This distinctive recognition differentiates C21.48A1 from antibodies directed against determinants accessible on HLA-associated B2M. Beta-2 microglobulin is a small protein that normally associates noncovalently with major histocompatibility complex class I heavy chains and is essential to the structure and function of MHC class I complexes.

Within MHC class I complexes, B2M supports proper folding, stability and cell-surface expression of the HLA heavy chain. These complexes present intracellularly derived peptide antigens to CD8-positive T cells and thereby contribute to adaptive immune recognition and surveillance. Beta-2 microglobulin itself does not span the plasma membrane and can dissociate from the MHC class I heavy chain, producing free B2M in tissues and biological fluids.

Free and soluble beta-2 microglobulin are investigated in a variety of physiological and disease settings. Serum B2M levels reflect a combination of cellular production, turnover and renal clearance and may increase during immune activation, inflammation and infection. Elevated levels are also associated with several hematologic malignancies and renal disorders. Urinary beta-2 microglobulin is studied in relation to proximal renal tubular function and conditions affecting renal handling of the protein.

Beta-2 microglobulin is also relevant to cancer and immunology research through its relationship with MHC class I antigen presentation. Alterations affecting B2M expression or its association with HLA class I can influence immune recognition of cells. The ability to investigate different structural states or determinants of B2M can therefore complement studies using antibodies directed more broadly against beta-2 microglobulin and MHC class I-associated protein.

Clone C21.48A1 is a mouse monoclonal antibody generated using soluble beta-2 microglobulin as the immunogen. It recognizes a hidden B2M determinant exposed when beta-2 microglobulin is separated from the HLA heavy chain and has been evaluated by Western blot, immunofluorescence and flow cytometry in human cells.
