B7-H6 Antibody B7H6/4821 / NCR3LG1 Monoclonal Antibody recognizes B7-H6, an immunoregulatory cell-surface protein encoded by the NCR3LG1 gene. B7-H6 is a member of the B7 family and functions as a ligand for natural cytotoxicity receptor 3 (NCR3), commonly known as NKp30. Binding of B7-H6 to NKp30 provides an activating signal to natural killer (NK) cells and can promote cytotoxic activity and cytokine production against cells expressing the ligand.
B7-H6 is a type I transmembrane glycoprotein containing extracellular immunoglobulin-like domains. These extracellular domains mediate recognition by NKp30, linking expression of NCR3LG1 on a target cell with activation of NK cell effector functions. The B7-H6/NKp30 interaction is therefore an important component of innate immune recognition and has become a useful model for studying how NK cells respond to cellular transformation and stress.
NCR3LG1 has received particular attention in cancer research because B7-H6 expression has been reported in numerous transformed cell types and malignancies while showing more restricted constitutive expression in many normal tissues. This expression pattern can provide NK cells with a mechanism for recognizing potentially abnormal cells. Changes in B7-H6 abundance or accessibility at the cell surface may consequently affect the strength of NKp30-mediated immune recognition and influence susceptibility to NK cell cytotoxicity.
B7-H6 biology can also be regulated beyond its expression at the plasma membrane. Cellular stress and inflammatory signaling can influence NCR3LG1 expression, while release of soluble B7-H6 can alter interactions with NKp30. These mechanisms have made the protein relevant to studies of tumor immune surveillance, immune evasion, inflammatory signaling, and regulation of innate immune responses. The alternative names NCR3LG1, B7H6, and NKp30 ligand reflect the protein's gene designation and its functional relationship with the NKp30 receptor.
