HIER was performed by boiling tissue sections in pH 9 10mM Tris with 1mM EDTA for 20 min followed by cooling before testing. Strong brown staining is broadly distributed through the neuropil and cellular processes, with nuclei remaining comparatively unstained. The widespread cytoplasmic pattern supports tissue detection of ARF1 with the ARF1 Monoclonal Antibody.

Purified ADP-Ribosylation Factor 1 Antibody ARF1/2117 SDS-PAGE. SDS-PAGE analysis of 2 ug purified, BSA-free ADP-Ribosylation Factor 1 Antibody clone ARF1/2117 under non-reducing (NR) and reducing (R) conditions. The non-reduced preparation shows a predominant intact immunoglobulin band near 150 kDa, while reduction produces the expected antibody heavy and light chain components near 50 kDa and 25 kDa. ADP-Ribosylation Factor 1 Antibody ARF1/2117 / ARF1 Monoclonal Antibody recognizes ARF1, a small GTP-binding protein belonging to the ADP-ribosylation factor family within the Ras superfamily. ARF1 functions as a molecular switch by cycling between GDP-bound inactive and GTP-bound active states. This regulated cycle allows ARF1 to coordinate membrane trafficking, vesicle formation and maintenance of intracellular organelle structure, particularly within the Golgi apparatus and secretory pathway.

Activation of ARF1 is controlled by guanine nucleotide exchange factors, while GTPase-activating proteins promote conversion back to the GDP-bound state. Active ARF1 associates with cellular membranes and recruits coat proteins and other effectors required for cargo transport. A particularly well-characterized function is recruitment of COPI coat components to Golgi membranes, facilitating retrograde trafficking within the Golgi and between the Golgi apparatus and endoplasmic reticulum.

ADP-Ribosylation Factor 1 also contributes to membrane remodeling through regulation of lipid-modifying enzymes and membrane composition. These activities connect ARF1 with cargo sorting, Golgi organization and the formation of membrane environments required for intracellular transport. Through interactions with multiple effectors, ARF1 can coordinate changes in membrane structure with movement of proteins and lipids between cellular compartments.

ARF1-dependent processes also intersect with pathways involved in cell proliferation, metabolism and cytoskeletal organization. Altered regulation of ARF1 has therefore been investigated in cancer and other disease models involving changes in intracellular trafficking or cellular signaling.
