Description

APEX1 Antibody / AP Endonuclease Antibody recognizes apurinic/apyrimidinic endonuclease 1, a multifunctional DNA repair and regulatory protein encoded by the APEX1 gene. APEX1 is a major component of the base excision repair pathway, where it processes apurinic/apyrimidinic sites created by spontaneous DNA damage or following removal of damaged bases by DNA glycosylases. This activity is important for maintaining genome integrity and limiting the accumulation of potentially mutagenic DNA lesions.

APEX1 cleaves the phosphodiester backbone immediately adjacent to an abasic site, generating an intermediate that can be further processed by DNA polymerases and ligases. Through this AP endonuclease activity, the protein contributes to repair of oxidative, alkylation-associated and endogenous DNA damage. APEX1 also possesses additional DNA-processing activities that contribute to the maintenance and repair of damaged DNA.

In addition to its enzymatic role, APEX1 is historically known as Ref-1 because of its redox regulatory function. This activity can influence the DNA-binding capacity of several transcription factors, connecting cellular redox conditions with transcriptional regulation. The combination of DNA repair and redox functions has made APEX1 an important target in studies of oxidative stress, genome maintenance and cellular responses to DNA damage.

Altered APEX1 abundance, localization and activity have also been investigated in cancer. Increased DNA repair capacity may influence cellular responses to genotoxic stress, while the regulatory functions of APEX1 can affect transcriptional programs associated with cell survival and proliferation. These properties have generated considerable interest in APEX1 as a molecular target in cancer biology and therapeutic-response research.

Immunogen

An amino acid sequence from the middle region of human APEX1 (RGLVRLEYRQRWDEA) was used as the immunogen for this APEX1 antibody (100% homologous in human, mouse and rat).

Storage

After reconstitution, the APEX1 antibody can be stored for up to one month at 4oC. For long-term, aliquot and store at -20oC. Avoid repeated freezing and thawing.
