APC Antibody rEMM43 Staining of Human Testis IHC. Immunohistochemistry testing of FFPE human testis with APC Antibody clone rEMM43 shows prominent brown cytoplasmic staining in germ cells within the seminiferous tubules, with comparatively limited staining in surrounding stromal tissue. Nuclei are counterstained blue. The inset PBS-only negative control shows minimal background signal, supporting the specificity of the observed Adenomatous Polyposis Coli Antibody staining.
APC Antibody rEMM43 Purity Analysis by SDS-PAGE. SDS-PAGE analysis of purified recombinant APC Antibody clone rEMM43 under reduced (R) and non-reduced (NR) conditions with 2 ug antibody loaded per lane. Under reducing conditions, the antibody resolves into bands corresponding to the immunoglobulin heavy and light chains at approximately 50-55 kDa and 25 kDa, respectively. Under non-reducing conditions, the intact antibody migrates at approximately 150-160 kDa, supporting the expected molecular integrity and purity of the recombinant monoclonal antibody preparation.

APC Antibody rEMM43 / Adenomatous Polyposis Coli Antibody recognizes APC, a large tumor suppressor protein encoded by the APC gene. Adenomatous polyposis coli is a central negative regulator of canonical Wnt signaling and has important functions in epithelial homeostasis, cell polarity, migration and cytoskeletal organization. Loss of normal APC function is strongly associated with colorectal tumorigenesis and inherited familial adenomatous polyposis.

APC controls Wnt signaling largely through regulation of beta-catenin. The protein participates in a destruction complex with AXIN, GSK3 and other regulatory proteins that promotes phosphorylation and subsequent degradation of beta-catenin. When APC function is lost, beta-catenin can accumulate and drive transcription of Wnt-responsive genes involved in cellular proliferation and differentiation. APC mutations therefore provide a direct molecular connection between disruption of Wnt pathway control and the development of colorectal neoplasia.

Beyond beta-catenin regulation, APC interacts with components of the microtubule and actin cytoskeleton and contributes to cell migration, adhesion and maintenance of cellular architecture. The protein can associate with growing microtubule ends and is found at several cellular locations, including the cytoplasm, cell membrane, adherens junctions and cytoskeleton. These functions demonstrate that APC acts as both a signaling regulator and an organizer of cellular structure.

Alterations of the APC gene are particularly important in colorectal cancer. Germline pathogenic variants cause familial adenomatous polyposis, while somatic APC alterations occur frequently during sporadic colorectal tumor development.
