CD68 Antibody SPM130 / Mouse Monoclonal recognizes CD68 using the mouse monoclonal clone SPM130. CD68 is a lysosome-associated transmembrane glycoprotein that is highly expressed by macrophages, monocytes and other cells of the mononuclear phagocyte system. As one of the most widely used macrophage markers, CD68 is routinely employed to identify tissue macrophages, evaluate inflammatory infiltrates and investigate innate immune responses in both healthy and diseased tissues. CD68 is localized primarily within lysosomes and endosomal compartments, reflecting the specialized phagocytic function of macrophages. Tissue-resident CD68-positive macrophages are abundant in organs including spleen, liver, lung, lymph node and brain, where they participate in pathogen clearance, antigen processing, tissue remodeling and maintenance of immune homeostasis. CD68 Antibody, clone SPM130 is well suited for studies examining macrophage localization, activation and distribution in both normal tissues and experimental disease models.
Altered CD68 expression is frequently associated with chronic inflammation, autoimmune disorders, infectious disease, fibrosis, neurodegeneration and cancer. Quantification of CD68-positive macrophages is commonly used to assess inflammatory burden, characterize tumor-associated macrophages and monitor changes within the tissue microenvironment. The optimal dilution of the CD68 Antibody SPM130 for each application should be determined by the researcher.
1. Staining of formalin-fixed tissues is enhanced by boiling tissue sections in pH 9 10mM Tris with 1mM EDTA for 10-20 min followed by cooling at RT for 20 minutes.
2. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.
A human alveolar macrophage subcellular fraction was used as the immunogen for this anti-CD68 antibody.
Store the anti-CD68 antibody at 2-8oC (with azide) or aliquot and store at -20oC or colder (without azide).
