AFF4 Antibody Human Placenta IHC. Immunohistochemical staining of FFPE human placental tissue using AFF4 Antibody / AF4/FMR2 Family Member 4 Antibody at 2 ug/ml. Heat-induced epitope retrieval was performed using pH8 EDTA buffer, followed by HRP-conjugated secondary antibody detection and DAB chromogen. Predominantly nuclear staining is observed in placental trophoblast cells, consistent with the expected localization of the transcriptional regulator AFF4. This AFF4 Antibody, also called AF4/FMR2 Family Member 4 Antibody, is suitable for studies of transcription elongation, Super Elongation Complex biology, nuclear protein expression, and gene regulation.
Western blot testing of human 1) HeLa, 2) HepG2, 3) 293T and 4) U-87 MG cell lysate with AFF4 antibody. Predicted molecular weight ~127/98/39 kDa (isoforms 1/2/3).
Flow cytometry testing of fixed and permeabilized human SiHa cells with AFF4 antibody at 1ug/10^6 cells (blocked with goat sera); Red=cells alone, Green=isotype control, Blue= AFF4 antibody.
Immunoprecipitation of AFF4 protein from 500ug of human 293T whole cell lysate with 2ug of AFF4 antibody.
AFF4 Antibody Mouse Brain IHC. Immunohistochemical staining of FFPE mouse brain tissue using AFF4 Antibody / AF4/FMR2 Family Member 4 Antibody at 2 ug/ml. Heat-induced epitope retrieval was performed using pH8 EDTA buffer, followed by HRP-conjugated secondary antibody detection and DAB chromogen. Predominantly nuclear staining is observed throughout the brain parenchyma, consistent with the expected localization of the transcriptional regulator AFF4. This AFF4 Antibody, also called AF4/FMR2 Family Member 4 Antibody, is suitable for studies of transcription elongation, nuclear protein expression, Super Elongation Complex biology, and gene regulation.
AFF4 Antibody Rat Brain IHC. Immunohistochemical staining of FFPE rat brain tissue using AFF4 Antibody / AF4/FMR2 Family Member 4 Antibody at 2 ug/ml.
AFF4 Antibody Human, Rat and Mouse WB. Western blot analysis of lane 1: human HeLa, lane 2: human HepG2, lane 3: human 293T, lane 4: human U87, lane 5: rat brain, lane 6: rat C6, lane 7: mouse brain, and lane 8: mouse Neuro-2a lysates using AFF4 Antibody / AF4/FMR2 Family Member 4 Antibody at 0.5 ug/ml. A predominant band is detected at approximately 150 kDa across all samples, although the predicted molecular weight of AFF4 is approximately 128 kDa. The slower migration is consistent with the reported behavior of this large nuclear protein and may reflect post-translational modification or anomalous electrophoretic mobility. This AFF4 Antibody, also called AF4/FMR2 Family Member 4 Antibody, is suitable for studies of transcription elongation, Super Elongation Complex biology, nuclear protein expression, and gene regulation across multiple species.

Description

AFF4 Antibody / AF4/FMR2 Family Member 4 Antibody recognizes AFF4, a nuclear transcriptional regulatory protein that functions as a central scaffold of the Super Elongation Complex (SEC). AFF4 coordinates the assembly of multiple transcription elongation factors, including positive transcription elongation factor b (P-TEFb), ELL proteins, and additional SEC components, enabling efficient RNA polymerase II-mediated transcriptional elongation. Through this scaffolding function, AFF4 plays an essential role in regulating gene expression programs required for normal cellular growth, differentiation, and development. NSJ Bioreagents provides AFF4 Antibody / AF4/FMR2 Family Member 4 Antibody for researchers studying transcriptional regulation and chromatin-associated protein complexes.

AFF4 is predominantly localized within the nucleus, where it facilitates the transition of RNA polymerase II from promoter-proximal pausing into productive transcription elongation. The Super Elongation Complex is particularly important for genes that require rapid and tightly regulated expression, including developmental regulators and stress-responsive genes. Because AFF4 organizes multiple SEC components into a functional complex, disruption of its expression or activity can influence broad transcriptional networks and alter cellular responses to developmental and environmental signals.

Interest in AFF4 has expanded because abnormal transcription elongation contributes to cancer, developmental disorders, and viral gene expression. AFF4 and other SEC proteins have been implicated in leukemias involving mixed-lineage leukemia (MLL, also called KMT2A) rearrangements, where aberrant recruitment of the SEC promotes dysregulated transcription of oncogenic target genes. AFF4 has also been investigated in studies of embryonic development, stem cell biology, and host-pathogen interactions, making it an important target for understanding mechanisms of gene regulation and transcriptional control.

An AFF4 Antibody is a valuable research tool for investigating AF4/FMR2 Family Member 4 expression, Super Elongation Complex assembly, RNA polymerase II transcription elongation, chromatin-associated protein interactions, and transcriptional regulation in normal physiology and disease.
