APC Antibody ALi 12-28 / APC Tumor Suppressor Antibody recognizes an N-terminal region of adenomatous polyposis coli (APC), a large tumor suppressor protein with a central role in regulation of canonical Wnt signaling. APC helps control cellular beta-catenin levels and contributes to normal epithelial homeostasis, cell adhesion, migration and cytoskeletal organization. Disruption of APC function is strongly associated with colorectal tumorigenesis and familial adenomatous polyposis.
APC is a core component of the beta-catenin destruction complex, where it works with AXIN, GSK3 and other proteins to promote beta-catenin phosphorylation and degradation. Loss or truncation of APC can disrupt this regulatory process, allowing beta-catenin accumulation and increased transcription of Wnt-responsive genes. These effects help explain why APC alterations are among the most important molecular events in colorectal cancer development.
Clone ALi 12-28 is a well-established mouse monoclonal antibody directed against the N-terminal portion of APC. Epitope-mapping studies localized ALi 12-28 recognition to a region encoded by exons 2-3, making the clone useful for detecting APC forms that retain this N-terminal sequence. Published studies have used ALi 12-28 to investigate endogenous and truncated APC proteins, including experiments in colorectal cancer models.
The N-terminal recognition of this APC Tumor Suppressor Antibody is particularly relevant because many cancer-associated APC mutations generate C-terminally truncated proteins while retaining N-terminal sequences. ALi 12-28 has been used experimentally to detect truncated APC proteins that are not recognized by antibodies directed against lost C-terminal regions.
