Western blot analysis of 6xHis Tag Antibody in recombinant protein samples. Multiple epitope-tagged recombinant proteins containing V5, FLAG, HA, and His sequences were resolved by SDS-PAGE under reducing conditions and probed with 6xHis Tag Antibody clone 6HIS/13239R. Distinct immunoreactive bands are observed at approximately 18 kDa and 14 kDa, corresponding to 6xHis-tagged STAR and 6xHis-tagged tryptase proteins, respectively, as indicated. The selective detection of the His-tagged constructs demonstrates specific recognition of the His epitope tag by clone 6HIS/13239R without cross-reactivity to non-His epitope tags present in the samples.

Description

6xHis Tag Antibody recognizes the hexahistidine sequence, also known as the His epitope tag, a short polyhistidine motif engineered into recombinant proteins for purification and detection. 6xHis Tag Antibody, also referred to as His Epitope Tag antibody, His Tag antibody, and anti-6xHis antibody in the literature, detects histidine-tagged fusion proteins expressed in bacterial, mammalian, or insect systems. Clone 6HIS/13239R is produced as a recombinant rabbit monoclonal antibody, providing defined specificity and consistent performance for detection of 6xHis-tagged recombinant proteins in research applications.

The His epitope tag typically consists of six consecutive histidine residues positioned at the N-terminus or C-terminus of a target protein. This small sequence enables purification using immobilized metal affinity chromatography and facilitates immunodetection in downstream analytical workflows. Because the 6xHis tag is compact and generally does not disrupt protein folding or biological activity, it remains one of the most widely used affinity tags in molecular biology and recombinant protein engineering.

Detection of His epitope-tagged proteins is critical for confirming expression, verifying purification efficiency, and assessing molecular integrity. Under denaturing conditions, intact fusion proteins are typically detected at their predicted molecular weight, while truncated or partially degraded forms may also be observed depending on construct design and expression system stability. Accurate recognition of the hexahistidine epitope therefore supports reliable interpretation of recombinant protein experiments.

Clone 6HIS/13239R recognizes the 6xHis sequence independently of the fused protein backbone, supporting broad detection across diverse recombinant constructs. The recombinant rabbit monoclonal format promotes lot-to-lot consistency and defined epitope recognition, enabling reproducible detection of His epitope-tagged proteins across experimental workflows.

6xHis Tag Antibody clone 6HIS/13239R is suitable for research applications focused on detection, characterization, and analysis of recombinant proteins containing a His epitope tag.

Application Notes

1. Optimal dilution of the 6xHis Tag/His Epitope Tag antibody should be determined by the researcher.

2. This 6xHis Tag/His Epitope Tag antibody is recombinantly produced by expression in CHO cells.

Immunogen

Hexa-histidine tagged human CD70 recombinant protein was used as the immunogen for the 6xHis Tag/His Epitope Tag antibody.
