Description

This antibody specifically reacts with an ~ 58kDa protein identified as Vimentin. It shows no cross-reaction with other closely related intermediate filament proteins (IFP’s) such as desmin, keratin, neurofilament, and glial fibrillary acid protein. Vimentin antibody alone is of limited value as a diagnostic tool; however, when used in panels with other antibodies, it is useful for tumor sub-classification. Expression of vimentin, when used in conjunction with keratin antibody, is helpful when distinguishing melanomas from undifferentiated carcinomas and large cell lymphomas. All melanomas and Schwannomas react strongly with vimentin antibody. It labels a variety of mesenchymal cells, including melanocytes, lymphocytes, endothelial cells, and fibroblasts. Non-reactivity with vimentin antibody is often considered more useful than its positive reactivity, since there are a few tumors that do not contain vimentin, e.g. hepatoma and seminoma. Vimentin antibody is also useful as a tissue process control reagent.

Application Notes

The concentration stated for each application is a general starting point. Variations in protocols, secondaries and substrates may require the antibody to be titered up or down for optimal performance.

1. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes.
2. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.

Immunogen

Recombinant protein was used as the immunogen for the Vimentin antibody.

Storage

Store the Vimentin antibody at 2-8oC (with azide) or aliquot and store at -20oC or colder (without azide).
